I think my most recent nope moment, without giving spoilers, was in Valdis Story, near the end where stuff is happening and you have to run..and I miss a ledge..and then a second time. That game is awful..ly fun!
Planty of hard moments in games:

Fade to Black - jumping over moving forcefields in a passage (you could see them only wearing some kind of glasses)

Wing Commander 3 - mission with Skipper Missile and of course final mission (I cheated in this one)

Starlancer - last mission with rescuing Steiner. I was really frustrated about this one. When I was swearing AGAIN my wife asked me with her calm voice: why are you playing sth that instead giving you fun and relaxation gives you only bad nerves...Wrrr

Shadow of the Horned Rat - the first 'serious' mission with orcs when they hold Carlson's Family.

Oh, god, yes! That was a huge NOPE moment for me, as well.

And Valdis Story is a superb game, by the way, if you're any sort of metroidvania / platformer fan, go get it if you haven't already.
Pokemon Heartgold- Picture this: Its my Sharpedo against my friends Slowbro. I was faster, I thought I hit crunch... but it was whirlpool. WHIRLPOOL. I don't even know why it knew that move. It was hard containing my rage as poor Sharpedo was killed by the Slowbro. It didn't help that we were having the battle in a french lesson.

This is what occurred to me immediately. I got down there. started the battle, then... nope. Gonna reload and go away for a while.

Also the first time I ran into Firkraag and thought "Oh, I can take him down". Nope.

Even better, there's a mod which makes them non-aggressive.

I'll agree with that.

Also, the RPG Maker game. One Night, the monsters in the final bit. They are so much more random and I went in only able to take one more hit. I didn't enjoy the length of time it took to get through that. Would prefer to punch myself in the face instead.

Maybe turning down the difficulty might help.
Ooh just thought of an old school nope : The first Golden Axe
The first time you enter the castle and just before the throne room. By the time I had got there I might have had 1 life left and I remember walking past some incredibly fierce looking green statues against the wall. I remember I was incredibly scared that they might come to life and utterly waste me.

Another 'nope'. Falling on to the ground before Death Adder and anticipating his barrage of spells...

Damnation! I can't find a single screenshot of that part of the castle (its just after the bottomless pit section with all the skeletons). If I have time tomorrow I'll try to get some screengrabs myself, with the dwarf naturally...

Never have I regretted an arcade game more. Think I pumped $20 in quarters in one day and still couldn't beat it. Though I will admit it was maybe the third time my friend and I played. Well, maybe X-Men and Turtles also.

No its not hard just very creepy the sounds and the way it walks.
