NPCs, dialogue and romances have not been bested.

Please tell me you're negotiating!

lochok: I can't believe nobody has mentioned Grim Fandango! Without any doubt (in my mind) the greatest game ever made... I would love a remix version - same game with re-rendered (or not pre-rendered) graphics which would look fantastic at modern resolutions - like they did with RealMyst or something....

My favourite game of all time too. I can't stand remakes, but I think a graphical revision mod of the game would be a great idea.
Half Life 2 would be my only other fanboy game. I'm a big fan of the series and think they are the best FPSs ever made.
Dungeon Keeper.
And Dungeon Keeper 2.
Now I've moved to Vista 64, I can no longer get DK1 to run at all and DK2 crashes a lot.
I pine for these games, I waste away with longing.
I've tried to get DK1 working with DosBox, but get an obscure 'cannot change resolution' error message that I can find no answer to, even after scouring the Internet.
Life is indeed a bitch
Legacy of Kain series and Silent Hill series.
These games ate a lot of my time...
Starsiege Tribes without a doubt. There is no other game out there that I have ever played somewhat consistently throughout the last 10 years. Sadly though my days of Tribes are over thanks to Vista. I hope to one day see Tribes on GOG.
(Runner Up - Half-Life).

Super Mario Brothers, best game ever made
