Hmmm... could it be that it turned into a lump of coal? Those can be hard to run on modern machines.

Just did, didn't do anything .

Make sure you clean off everything,Run CCleaner and any other Registry Cleaners.
Also delete leftover files on drive.

Did you change/update anything between when the game was working and when it started giving that error?

Are you shutting down all non-essential programs (especially anti-virus) before starting the game? Firewalls have also caused problems on startup, but not that error, that I've seen reported before.

Try starting the game directly from the '..\Divinity - Original Sin\Shipping\EoCApp.exe' program file, by right clicking and running as administrator. Also, you could try right clicking, selecting Properties and switching to the Compatibility tab (default shortcut for me in Win 7 64 has run as administrator and disable display scaling on high DPI settings checked).

Try browsing to the '..\Documents\Larian Studios' folder and renaming the 'Divinity Original Sin' subfolder.
This folder contains the saved games, configuration files and a level cache folder. The game should recreate it on startup the next time it is run (aside from the saves and the DLC subfolder, which the GOG version uses to keep track of the companion and (if applicable) Source Hunter DLC).
If that lets you start the game and create a new profile, exit, maybe reboot and make sure you can restart the game. If so, move the saves from the renamed folder into the new profile's savegames folder, and move the DLC folder to the new D:OS folder.

Yes one of those could be a problem although never had a problem V.P.I know from experience Windows (any)can be a pain in the arse when trying to get games running smoothly.I would delete game icon on desktop and go into the files and see how many EXE.FILES and try them.It certainly can't hurt anything.I remember Anno 1404 Venice,couldn't get it to kick so I deleted desktop icon.In the files I found (2)game launchers and tried them first one failed 2nd one worked perfectly after that.When game was installing it sent the wrong launcher or icon to desktop..

No idea what happened, but I just tried opening it again today, and it's working :P

The only thing I had done different between the last time I had played and the time it stopped working was installing 2 other games, but nothing else . Anyways, it's working now, so I guess the only thing I had to do was restart my computer :P

Every time you install a game, it's better to install all included runtime libraries within installer (e.g. VC++ runtimes) and RESTART your system, even if installer does not suggest it.

Also, a "clean install" is helpful:
1. Uninstall
2. Delete game folder and game data folders (usually it's on windows drive)
3. Clean registry (there maybe leftovers by game)
4. Clean temporary folder (TEMP folder)
5. Restart
6. Reinstall the game + provided runtimes
7. Restart
8. Play!
