The crown that you got from the albino rat is cursed. Your screen flashes, when you put it on.

Considering the mani rune in the secret room: Wen you create a mage or a druid, you'll always get the rune. A paladin doesn't always get it. I created a paladin with an int of 22 and no casting skill and I got a shortsword. Then I made a paladin with an int of 17 and a casting skill of 8 (which happens to be my highest skill together with defense, no idea whether that's relevant) and I got a mani rune instead of the shortsword.

But lesser healing is a spell of the second circle, so you'll need to be at least level 3 or 4 to actually cast it.
