Simply I click on her to star the mission in the Estherian Ruins but after she talks, I give the OK but nothing happen and the question mark remains above her head!!!

It's a known bug/is there a solution?

I think you have to step on the portal to activate it. If I remember correctly, the difficulty level increases from left to right (but they're not all that different in power level)

The problem is that the mission is not starting. After I clicked OK she's not doing the animation and she remains with the "?" on her head!

Try reloading the level by traveling back to Torchlight (via the waypoint portal one stair above or via scroll) and returning to the Estherian Ruins.

Done and nothing changes, I also tried to do another mission and then come back but the problem remains.

You have to find the glyphs in the portals, and my problem is that the cutscene in which Sly opens the three portals is not starting!

Hmm, looks like the quest doesn't trigger like it should. Probably you are missing a requirement.
You should try going back up one level and kill the Overseer again. Make sure, that there're no enemies left on this level. If it's already clear, wait until the monsters re-spawn (don't know how long that takes) by doing other levels (e.g. buy some maps in Torchlight).

I simply went back and did the Overseer level backwards until I defeated all the enemies.
When I went down the stairs it played again the cutscene of the book and then the mission started!!!
