In my opinion, there's only one true way to build each class. The trick is to determine how both synergies synergize with each other and the skills that benefit others, then build around it. I've used the online skill calculator to figure out what I think is the ideal way to go.

For example, any Defense class needs to use a shield in melee combat, focusing primarily on physical damage. Classes made with Warfare focus on physical damage. Classes using Nature rely on wolves for distraction and dealing damage.

I've seen a lot of Icefrzzy's videos on their channel where they use multiple builds for each class. Most classes they make that use Hunting go either bow or spear route, but I figure that with enough extra skill points from equipment, you can go both ways. They even made a Conqueror using dual wield, but it seems a waste of a mastery since many skills require a shield.

For certain classes, the way to build is obvious. To me, a Spellbinder would be better off using a shield and fighting in melee combat inflicting physical damage as there's nothing in Defense that could benefit the vitality damage of certain skills in Spirit. Battlemages are better off using physical damage because nothing in Warfare affects fire and burning damage. Guardians fight in melee with a shield together with wolves as do Champions with dual wielding, using wolves to fight for and with them, their own damage buffed by them.

What do you think? How many different ways do you think classes can be made? Should I focus only on one build per class?
That's entirely up to you and how much you enjoy each class. That's the beauty of the game, that you can build classes in different ways.

The list was apparently made before any of the dlcs released after Immortal Throne since they don't use the increased mastery bar, level caps, or include builds using Rune or Neidan.

So if I wanted to create a class using Hunting for example, I need to develop a build around the use of the bow or spear, but not both? Or with a class with Nature, I can be someone who inflicts damage on my own with pets supporting me, or relying on pets to do all the damage, but not both?
Yes, it was made many years, but I think the basics still are good.

Depends on how many points you need in your skills.

When I play Hunting it's always primarily a bow for me (maybe throwing weapons instead), but I often have a spear on the 2nd weapon slot for close in fighting.

And as said in your other thread, pets are usually all pets so you're a petmancer or they act as support while you do the main damage.
I spent much of yesterday making a list of all the builds I could come up with using the online skill calculator at https://bytesquire.github.io/TitanQuestCalculator/mods/Vanilla/index.html

Some classes only have two that I could think of and others have up to five. I came up with a total of 148 at last count.
I'm currently reviewing the list and removing any builds that wouldn't work out or don't properly synergize each class. I would think that the Hunting mastery requires your character to use a bow, spear, or throwing weapon, and making an elemental caster using either Earth or Storm would be a waste of a mastery since the majority of the skills in Hunting only work with certain weapons.

Defense mastery has many skills that require a shield and it would be a waste to make a character that dual-wields or uses a staff.

Storm is primarily an elemental mastery with nothing to benefit physical damage, so physical characters become hybrids. And even those using Earth mastery end up having some fire damage even with just one point in Earth Mastery, making more hybrids.

Certain classes make better warriors, casters, etc than others..
