Try activating Vsync, there were issues with ladders when it's deactivated.

Climbing ladders usually is a simple matter of moving into them. You'll automatically start looking up and can use the forward/backward movement keys to climb.

Hi, It is activated, so I disabled it and then enabled it and I've still got the same problem.

Did you enable it in the game menu or at your graphics card control panel? Try enabling it in both.
Not sure how to check if Vancouver is turned on/off on the video card, I have looked but can't seem to find where. I am also having issues with lock picking not starting at locked doors/chest ect.
I really don't like auto correct, Vancouver was supposed to be Vsync.

Depends on your graphics card's control panel.

If you're using the Catalyst Control Center look under 3D settings for a slider named "Wait for vertical refresh". There you can turn V-sync on.

In Nvidia's Control Panel it's also under 3D settings. Look for Global Settings and scroll down to "Vertical sync". There you can turn it on. (If it works, you could turn it off in Global Settings again and turn it on in Program Settings only for the game, but I'd start testing with global settings, just to make sure the setting is applied).

You may have it already, but..

I found that sometimes I had to do a jump. Stand at the base of the ladder, hit jump then the move forward key.
I did a rebind on my keys so I move up to the ladder, click on space bar then press the s key and I move up.
Don't know if you really need to do that but for some reason I had to in order get climbing. The game has various buggy issues for me and this was one of them.
I am playing 'A Friend in Need' and have reached the Architects study. Cut him down and got the loot but cant climb the ladder to get to the map on the top floor!
