Try reinstalling. Did you replace the rebexe file or something?
I have the same problem on an Asus transformer Book t100.
It was playing fine but then two days ago suddently started asking for a cd.
I reinstalled many times, wiped the registry, and reinstalled to no avail.
And no, I am not using any other rebexe file. Just the one the installer created.
Can someone help?
If DllLocation or TooBigLocation (in the registry) are pointed to the wrong locations, you'll get this error. Dunno why yours would be wrong, but that's how you fix it.

I started getting this error after I did the "alternative" method to get RebEd to work. (I.e. I opened up Regedit and pointed the two labels - "DllLocation" and "TooBigLocation" - to my E drive, as in "E:\Rebellion".)

RebEd works fine, but Rebellion itself won't start because it says there isn't a CD.

I have a full copy of the game on my C drive and on my E drive (an external USB hard drive). What do I need to do to get it working? Can I just edit the regedit back to point to the C internal directories?

Yes, you can. Or check out my tutorial on fixing RebEd so you can launch the game and have RebEd working simultaneously.
