Since the Space Quest games are now on a activision sale (still kinda hurts to connect Space Quest to Activision). I was wondering.

The 2006 collection as well as the steam version are unable to use General Midi for the music. While it isn't as good as a MT-32, it's still leaps beyond FM synthesis.

My question is simple, will the two collections be able to use General Midi for playback? or am I bound to FM (Adlib) like in the Steam and retail versions?

My question was already answered in the setup utility topic.
I'm glad you got your questioned answered; but I wanted to cover the topic in detail since a lot of people give up before getting things perfect.

sq4 and later support general midi, and sq3 supports mt-32 hardware

You need to edit the RESOURCE.CFG file in each game's installed directory
and change the line that says

soundDrv = ADL.DRV

to

soundDrv = MT32.DRV

or

soundDrv = GENMIDI.DRV

later SCI games will also have a audioDrv section; this is for digitized effects.

You may need to set the midiconfig line equal to your midi device's value if you have more than one midi port available, you can get a list of how dosbox enumerates it by using "mixer /listmidi" in a dosbox window. You can also use a software general midi program (I use coolsoft virtualmidisynth with the fluidR3_gm soundfont) or an MT-32 emulator.

GOG configured sq1 and sq2 to use the pc speaker; the games are capable of tandy 3-voice sound. You can enable this functionality by editing dosboxSQ1.conf (or dosboxSQ2.conf) and changing

machine=svga_s3

to

machine=tandy

Pretty much all of this also applies to the king's quest games as well.

This is great, but the step that is missing (for me anyways) is that I still need to get into the game setup menu to tell it to use my mt32. Unfortunately, when I click on the .exe file, my Win 10 machine less than politely tells me that my machine can't run that file. So I'm back to where I started, in that I have no way to play the game using the mt32.
Yes, you can't run the installer from Windows 10 because Windows won't run anything that's less than 32 bit. That's why you go into the resource.cfg file & adjust the setting in there. Or, if you can't figure out how to get it to work by editing the resource file, then just use ScummVM and set the audio to use the MT-32
