Anyone know how to set up overwatch in the Director's Cut version of Dragonfall? It allows me to choose where to "face" my character, but I don't know how to "confirm" overwatch.

Once you've rotated your character to the direction you want, simply click the mouse button. In other words, pretty much the same as before... :)

I've noticed DC seems a tad less responsive to mouse clicks sometimes - typically I have to click multiple times to get my guys to reload their weapons...

Hmm... for some reason, if you have the "double click" option on, it seems that you can't actually confirm the overwatch (That's what was happening to me originally). Once I switched to "single click" mode again, it fixed this issue.

Really? Never noticed since I never use double-click - did you mention this on the Harebrained forums? It sounds like a bug that they don't know about yet...
