Do we have to complete the whole "vanilla" game before unlocking this mode or is there another way?

Are you talking about the pixel level, by any chance?

You unlock it by obtaining 250 achievement points. As you keep playing through the game normally, you'll eventually get to that amount, as the points you get for finishing chapters should be enough to get you there.
