I have a question, if a walker accidentally slipped past a roadblock which I removed for a second when modifying my city into another part of the city, what then, am I screwed? Actually it happened both ways, a walker from part A went to part B, and a walker from part B went to part A.

If I block it again, will they despawn at some point or what? Keeping the road open only invites more walkers to slip.

It would be interesting to check. Roadblocks only block random walkers, not destination walkers. And a worker (that spreads the influence of his/her building?) is a random walker, but technically becomes a destination walker when he/she heads back to his/her building after having walked a set amount of tiles. So I'm pretty sure that no, the walker won't be stopped by a roadblock when heading back.

It should be easy to check. Build a long stretch of road with a temple or something, and then wait for the labour seeking dude to come out, then block the way back to the temple with a roadblock and check if the walker goes past the roadblock on his way back to the temple. These random walkers only walk a set number of tiles at a time before heading back.
Workers will ignore road blocks if returning to "Home Base" and they are on the back end of their journey.

Say an Architect has a route. He will walk in one direction for--- if I remember correctly--- 54 squares. The computer then will calculate the shortest distance back to home base. If that means the Architect needs to ignore road blocks, he will. You can actually use this to keep road blocks for other workers but still get an Architect to walk by even if it means he ignores the roadblock. The maximum distance varies for the Workers but I think Police, Fireman, and Architects have the same distance. The shortest route, again if I remember correctly, is by the Water Supplier.

Ofc that still means these workers will be useless for the remaining duration of their walks but at least all is not lost. Gotta be careful when messing with the roadblocks
There is a bug in the game where some workers simply ignore the road block altogether. Usually it is the girl from the market and you will see her in your industrial area because "She doesn't see no road block." I think it's funny, so if not too important, I'll leave it. But if it starts to effect the residential area, the only solution I have found is to raze the market and move it to a new location.
