When trying to cast "shrink monster" on the Troll, nothing happens.

Hi,
try to cast it while the troll is distracted with Diego. Keep the key you're using to cast the scroll pressed untill the troll shrinks. Don't let go of the key untill the troll is tiny :)
Also make sure that you're close to the troll. You should be close enough to see his name on the screen.

This only happens when I try to get the troll skin to make the ulu-mulu.

Are you alone or with Diego ?
If Saturas gave you the 'Search for the Focus Stones' quest, when you go to Focus Canyon (the place where the Troll is), Diego will be there to help you fight the goblins, the Troll and the Harpies that are guarding the Focus Stone.
Diego and the Troll are kind of 'immortal', so you just have to let them fight each other, approach the troll from behind, get close enough to see his name on the screen, and cast the spell.
I'm under the impression that in order for the spell to work, the troll can't be aware of you, i.e. attacking you.

By the time you get to the Ulu-Mulu quest you should be powerful enough to take down a troll without having to shrink it first. I'm not sure whether Shrink Monster actually works on trolls outside of the focus quest (I haven't tried it).

It does, I've tried it. It just takes ages for the scroll to charge up, so while it's possible to do it without Diego it's much harder in theory.
