spoderman: The Uncharted and Infamous series are my favorite exclusives, so definitely get them first.

F4LL0UT: Yeah, already ordered Infamous (should be here on Tuesday). Played it for an hour or two on a friend's console and it was pretty awesome, can't wait to give it a serious try. As for Uncharted - well, it can wait for now. I was very unimpressed when I tread it.
Which Uncharted did you try? I personally thought the 2nd Uncharted was the best, Uncharted 3 is amazing to look at, but not as fun to play. I'd rank them like this: Uncharted 2>Uncharted 1>Uncharted 3. If you didn't like the first, you might wanna try playing the 2nd one.

If you liked Infamous, you'll probably love Infamous 2, I thought it was the better game.. both are great though.

Let's see...

Valkyria Chronicles
Heavy Rain
3D Dot Game Heroes
Metal Gear Solid (get the whole series)
Catherine
Uncharted (if you want just one, the second is probably the best choice)
Red Dead Redemption
LittleBigPlanet (probably no need to get both, either one would work)

And grab EchoChrome off PSN.

Oh, and Final Fantasy X once the HD release is out.

Those are the ones I can vouch for myself.

Also there's a number of titles current and upcoming that I haven't played myself, but I find intriguing, and as such urge you to take a look at.

The Last of Us
Beyond: Two Souls
Journey
Unfinished Swan
Rain
The Unfinished Swan
Puppeteer

F4LL0UT: ...
Other than the games already mentioned i suggest the Jak and Daxter collection. It contains all the 3 games released on the PS2 with updated HD graphics.
Some of these aren't exclusives, but all worth getting never the less (in my opinion):

Ni no Kuni (an amazing game and there is a certain area that nulls the need to level grind for long hours once it's unlocked)
Tokyo Jungle (I think that's the name, it's available in english on PSN, or the Japanese version on disc)
Ico/Shadow of the Colossus Collection
Record of Agarest War series
Catherine (like others have said, it is an amazing game and very very frantic fun)
3D Dot Game Heroes (a fantastic game in the light of Dragon Warrior/Dragon Quest and Legend of Zelda)
Metal Gear Solid collection
Metal Gear Solid 4
Monster Hunter Portable 3rd HD Remaster (an import worth getting, and the work to figure out how to play it)
Enslaved: Odyssey to the West
El Shaddai: Ascension of the Metatron
Demon's Souls
Dark Souls
Doctor Who: The Eternity Clock (another import but worth the effort to get it)
Uncharted series

and one of the best games I've ever seen worth the effort to import it for any Dynasty Warriors type game fan...
Kamen Rider Battride War

It has just come out but it is friggin awesome! My copy hasn't arrived, but a friend has one and it is a blast. I recommend it to not only fans of Kamen Rider, but anyone who enjoys a really smooth and enjoyable 3rd person action game. :)

F1ach: Nobody mentioned Heavenly Sword, so check that out...pro tip : turn off the 6 axis controls. They also made Enslaved.
Yes, ninja'd me before I even posted! :D To the OP: Another vote for Heavenly Sword by Ninja Theory, better played before Enslaved. It is also directed by /starring Andy Serkis (aka Gollum), along with the sexy voice of Anna Torv (from "Fringe"). This was the first game I ever played on PS3 (a launch title in 2007), and although it is short and the graphics might look a bit crusty these days, the characters had me totally hooked throughout.

BTW, if you only played half way through Uncharted (1), you probably had not reached the part where the plot is revealed and the shit hits the fan... :)

Outland is an *excellent* platformer. It's kind of like a cross between Rygar and Ikaruga. It's a download only game available on PSN.

I've only made it halfway through the first Uncharted as well. I wasn't terribly impressed either, but the main reason why I haven't finished it is because I'm just not good enough of a game player to progress. I've essentially reached a point where I keep getting my butt handed to me over and over again. That's the one thing that I like about PC games so much more than consoles. Cheats are almost always available for PC games, whereas console games seem to rarely include cheat modes anymore. It sucks for us guys that enjoy the games but frequently stink at them.
I just picked up my PS3 at the beginning of the year and my recommendations would be:

Batman Arkham Asylum and Arkham City (The latter would be best if you only wanted one. GOTY edition of course.)
Castlevania: Lords of Shadow
Dragon's Crown (comes out in August)
Sonic's Ultimate Genesis Collection (40 games for $20 or less and a lot of RPGs)

I'll also third (fourth?) Folklore, ICO and the Shadow of the Colossus collection, and Dragon's Dogma: Dark Arisen.

If you're into hard but rewarding games: Ninja Gaiden Sigma.

EDIT: Anyone know of a good racing game on the PS3? I hated Midnight Club: Los Angeles, but I did like Forza 2 on the 360. I really didn't like the first two Gran Turismos so if they haven't changed, I probably won't like 5.

how the fuck did this thing nearly go two pages before someone recommended Journey?

It isn't a big game, or really an "Epic" game by today's standards, but Journey is pretty much a perfect game in that there is not a singe thread out of place in it.

*edit*

oh, yeah; get PS Plus as soon as you can.

it is like $50 for a year, or $18 for 3 months, but the free games are more than worth it.

even if you just get the three month you can instantly get Sleeping Dogs, Vanquish, Infamous 2, and a hand full of Inde games for free* each one of them is more than worth $18 on their own.

*some Bullshit applies, but it isn't as bad as Sony could be.

The most notable of the bullshit is probably that you loose access to any free games once you cancel your subscription to PlayStation Plus. That's why I don't bother with it. Actually owning games is one of the major advantages of consoles over most PC game retailers. I like to know that I will still be able to play the games years after the console is obsolete. (They do seem keen of giving that advantage up for the next generation) If that doesn't bother you, then go for it. Once upon a time I had a subscription to GameTap, so I do acknowledge how great a service PlayStation Plus really is. Its just not future proof.
Disgaea 4.
Unfortunately, 3 looks rather pixelated in HD. It's still good, but I enjoyed 4 more anyway.
In case if no one else recommended it, Ni No Kuni
