How do you gain access to the mission packs in Earth 2140. Do you have to complete all the missions in the base game before they get unlocked?

No, the mission packs are available from the start. There will just be other "buttons" for the mission packs in the main menu when you start the game. If you recall, in the menu for Earth 2140 there is the ED and UCS symbols you can click on to start their campaigns. Now, there are just a couple other buttons called "mission pack 1", etc.
Do note, however, that the AI in GoG's version of Earth 2140 does not build buildings or units, so they only have what they start with (IE its extremely easy, unlike the original, in which the AI was very tough).
