I barely started my first run, but from all the reviews I've read, you can expect at least 40h for a first run (maybe less if you're an exceptionally fast reader), probably a lot more if you like to take your time and being thorough (or save-scum to check different options in certain dialogues and skillchecks).
That being said, this is a game that begs for at least a second run, maybe more, to uncover everything it has to offer.
From what I understood, the conclusion of the investigation is more or less the same, but you just get there using different means based on the type of character you made (you distribute 12 points among four different primary abilities, each influencing a set of six secondary skills, including one "signature" ability).
