Okay, so having started up the game, I was getting the Microids logo, then nothing - it would simply stop and vanish - no task listed in task manager or anything.
So, I contacted support after fiddling with various compatability settings, and whislt I was going through various possible solutions with Konrad the support whiz, I had a brainwave: to add it to the list of "Data Execution Prevention" (DEP) exceptions.
Control Panel\System and Security\System - then click on "advanced system settings" on the left and under performance select "settings".
The third tab is "Data Execution Prevention". Click "add" and find the Amerzone exe file (most likely at C:\GOG Games\Amerzone by default), hit apply and you should be good to go!

I did have a further issue where the opening cinematic would play and then bring me back to the main menu. By unchecking all compatability settings in the shortcut except "run as administrator" and the "XP service pack 3" compatability mode, the game now runs fine.
My guess is the second problem is from me fiddling about with the settings earlier.
