Normally, I'm not into the whole DOSbox thing. But, with the special Interplay deal this week, I'm giving SS a good look. The thing I'm wondering about however is replayability. It seems like the kind of game that, once you've beaten it, there really isn't much reason to play it again.
I'm pretty sure there's no longer any multiplayer support for it so, are there any expansions or mods available to give it some longevity.

I don't think there are any mods but this version, the gog version, includes the Mission Pack.
Taken from the games wiki
"The game also includes multiplayer and a mission editor. Multiplayer can support up to 16 players, and a "single player anarchy" mode also includes options for co-op play, custom missions, bots, and playing single player missions with multiple humans, either with the aliens or against them."
So it sounds like you can fire up a skirmish type mode with bots and just have at in outside of the campaign. Also if you enjoy editing it should come with the mission editor.
Even if the game only keeps your intrest for the campaign its only $3 now with this interplay deal, so if its a game you think you would enjoy, now would be the time to pick it up.
