Is there a way to turn on the subtitles during the conversation? I'm not English and although I understand English pretty well I am unable to understand everything because of sometimes poor quality of speech and sometimes the dialect of the actor :(

Sadly I do not have this game yet so cant help you directly, but I've googled that subtitles are in one of files in game directory so there should be some way to turn them on. Have you tried run setup and disable sound for example?
Also I've found this video and it has some subtitles on (but maybe they were edited in).

I'm confused - there's an option in either Game or Audio controls to turn on text messages. Is that the same as subtitles? I get text for all conversations.

Yes, probably it is :) It seems that our friend did not run setup file at all just started to play at default settings.

Ah.. :( And again I can not help you directly because of the same reason. I was trying to google out the answer but nothing so far. :(

There are almost certainly no subtitles for LoL2, there is no ingame option to turn them on/ff :(

Well, I tried to find something but its hard without having the game installed. But it would be 1st thing to check for me when I get it during some sale, I hope it will be soon :)
