Jurassic Park Trespasser. Without a doubt the most professionally produced LP ever. Nothing else I have seen even comes close. It's also really informative, and good fun.

Shamus Young has really nice Let's Plays on his site. Only the Mass Effect one is a video (down at the bottom of the page), and it is ongoing. However, he uses stills in all of his Let's Plays, and I find his humor quite good.
He also has a Let's Play at The Escapist. Again, not a video, but it uses many screen-shots.
I know a lot of these are not videos, but they are just too good for me NOT to list them.

I am currently investigating that possibility, although there are several technical and logistical hurdles that need to be leapt over first. Will post updates to my situation on this thread at some point. No promises.
