Even a MMORPG in that universe is possible as it offers several fractions all hungering for power and with different interests.
That offers the possibility of quests.
The same goes on for Isaac Asimov's Foundation series and other series.
A lot of books are suitable to make an action, adventure or rpg from.
All that's needed is a good setting/universe, characters a player can bond with and a good story.

They did make Dune games. Westwood made Emperor: Battle for Dune and Dune II

Since the Thread has been resurrected anyway, I'd love to play an adventure game based on Tom Robbins' novel "Fierce Invalids Home From Hot Climates". There are too few 18+ humorous adventure games out there.
I wouldn't mind seeing a game made out of the Reality Dysfunction series by Peter F. Hamilton or the Otherland series by Thad Williams...
If I can add my 2 cents, I'd love to see an adaptation of the Cycle of Hyperion by Dan Simmons.
Maybe as a point and click adventure or a dialog-heavy RPG.
I'm getting monotonous, all i can talk about lately are adventure games :-D

I know of that game but unfortunately never managed to complete it.
Back than I was never able to find a game in stores in my area.
I've played a copied version and 1 from an abandonware site but at some point it kind of jammed and made it impossible to continue playing.
That version of Dune was partly based upon the movie Dune.
A new game based upon the books and less linear would be great.

That's what I get for not reading the entire thread :-).
I always had a soft spot for Dune1 - it was an awesome game... even if i had to upgrade my whole system to run the talkie version (installed a cd drive! soundblaster!! etc..)
It sticks pretty close to the book.
I guess most of the sci-fi and fantasy books i read could make good games... though in the translation to games they might all end up pretty generic.
Lets say:
Song of Ice and Fire. (though it looks like Dragon Age might do that for us...)
Ender's Game. (as a super-fast 3D RTS game with darwinia/defcon style graphics)
Jennifer Government (comedy near-future. bladerunner meets er... something)
Sophie's World (philosophical adventure game)
Tuesday Next novels (literary action/adventure in an alternate reality Swindon)
Lone wolf & cub (classic manga serial)
already been adapted, but:
Discworld RPG and RTS.
Dragonriders of Pern (a decent game)
More Judge Dredd and Rogue Trooper games (come on Rebellion!!)
I'm sure the books would suck now i'm grown up, but as a kid i loved the Belgariad novels, can't believe they haven't been made into a game yet.
Plus, how about the Gor novels? I'm sure there is a market for erotic fantasy games out there.... ;-)

A"To Kill a Mockingbird" lawyer game. Complete with Phoenix Wright style dramatic poses.
A Jack London book "The Sea Wolf" would make for a pretty cool point and click adventure.....

I've often wondered why there aren't shakespeare games, there's sex, bretrayal, violence, murder & good dialogue. Even a SCUMM engine style game would work, especially if you controlled multiple characters

I'm a huge Shakespeare fan and I can't believe I had never thought what they would be like in video game form. A point-and-click adventure game based off of Macbeth would be freaking sweet!
Also while were on the subject of point-and-click adventure games. They should definitely make some adventure games based off the best works of Jules Verne. They already made a spin off of The Mysterious Island. They should work the same magic with 20,000 Leagues Under the Sea and Journey to the Center of the Earth.
Hold on. I have to go faint in a pool of drool.
EDIT:
They should also make video games based on the Into Darkness book and its many sequels(Out of Darkness, Through the Darkness etc...). I haven't even read the books yet(Although I plan to soon) and I already know they would make great games. Imagine WWII with dragons and you get the idea.
