Due to patches like True Patch Gold or so, the autoexec.cfg might already exist. If so, just paste it into that.

- Start Vampire

I just tried it myself (with an xbox one controller) and it works fine. It looks like the profile needs some further tweaking, like setting up the movement keys, the use key... maybe other things?

edit: looks like the use key is mapped (combined with the reload key)
It looks like for some actions you need to control the mouse cursor with the right analog stick. This game wasn't made for controller, so it'll never have the 'complete' controller experience (like a little auto aiming).

before it did and it didn't work, so try the options of the partner above and nothing. With the Xpadder and now with DS4Windows in this moment nothing again
