I can confirm that it is a one time activation only.

You receive 2 keys (one for the base game the other for the DLCs) which you must activate on social.bioware.net using an origin online account. When the content is activated on you account the only thing remaining is to login to your origin account in the game.

It never asks you to re-login to your account, even if you go offline (the content should remain activated).
