Curses as in "from now on you'll turn into an ogre at night until my son... I mean prince charming rescues you or something"?
Shout out to Shrek 2.

In Hard West there's a character that becomes cursed. Or maybe it's 2 characters or a little more even. Lotta cursing going around that game.
From the topic of which you speak, I think, The Darkness. My real life curse is... Hmm.. To wish to be a writer and to know that I am destined as an invalid... As it was foretold. A fortune told to me that I would become that which I would find myself on my 35th birthday. On that day I found myself in the hospital.

The Madden Curse is the first one to come to mind...

Also, your new avatar pic is horrifying like a curse Fairfox. :P

Does Vampirism in TES Oblivion count?
Or is that more of a disease?

Well, Oblivion was bad but I'm not sure I'd call it a disease.

funny guy :P
In the Wizardry series:

Wizardry 4: Adept Baldness (there's also a key item that's cursed)

Wizardry 8: Bloodlust, Staff of Doom
Cursed items can't be unequipped. That might be mostly a rouge-like thing, but I played a lot of games where curses work that way.
I gave up trying to understand this topic, but anyway...

"Golgotha's Gift" in Lords of Magic used to give me fits, but now it's just a minor annoyance. It's much, much, much worse to have an enemy Shaman or Witch throw "Vortex" in the middle of my army.

For this, Baldur's Gate / Infinity Engine games / D&D titles come to mind. And they also use the Curse spell, essentially the opposite of Bless for combat pluses and minuses..

Real life curse? Lately I've been a deer magnet. After hitting one about nine days ago I've had four more close encounters, two of them in the same spot as the earlier collision. Probabilities catching up with me, maybe.
