edited out the prisoner problem...

And I've purchased the prime primer, and found the gun powder in the past... yet they don't show up, and there's no statue.

Do I need to actually finish a run to the end before things appear in the lobby?

There is an NPC on the fifth floor who you need to give those components to... so you need to acquire them and carry them down to her in a single run. Once you give her any specific component, you'll have it unlocked on that save file forever, so you only need to make it once! (Once per component that is...)
