Can this be played with one hand? I can alternate between mouse and keys, but not quickly.

For the first few levels perhaps, but you really need both hands on the mouse and keyboard to play once things pick up. This is one of those games where sometimes I wouldn't even dare scratch my nose so as not to risk my character getting killed.
